Download file to see previous pages Colorectal Cancer cells are aneuploid cells which undergo a large-scale chromosomal rearrangement. LOVO and LS-174T cell lines have KRAS mutations. LOVO lacks BAT- 26 locus. (Ahmed et al. 2013). According to Bu et al. (2011), the order of efficiency of cell lines is given as Caco-2> HT – 29 > LS174T > LOVO. LS174T cell line is best used for the investigation of the phenotypes. Similarly, the proliferative and invasive capacity of LS174T is very high than any other Colorectal Cancer cell line. Thus LS174T can be used to study mucin expression in colon cancer. (Bu et al. 2011). Bcl – 3 ( B –cell lymphoma 3 – encoded protein ) is an oncogene protein. It is a regulator present in the NF- kB. The studies on Bcl- 3 have found that Bcl- 3 is an essential oncogenic regulator for the study of Colorectal Cancer. Bcl- 3 is found to activate the homodimers such as p52 and p50. The phosphorylation of Bcl-3 results in upregulation. This is very important for colorectal carcinoma. (Puvvada et al. 2010).

CADCAM (Carcinoembryonic Antigen- Related Cell Adhesion Molecule) belongs to a group of the Carcinoembryonic antigen family (CEA). CEA contains two subgroups: CADCAM and Pregnancy-specific glycoproteins (PSG). CADCAM is found to be a tumour suppressor molecule regulating the growth and differentiation in the cancer cell lines. (Finkenzeller et al. 2003). There are six types of CADCAM. CAECAM1 and CAECAM6 act as biomarkers for the study of colorectal carcinoma. CAECAM6 is upregulated in the colon cancer cells and thus acts as the potential target for colon cancer treatment and diagnosis. CADCAM are the target for monoclonal antibody therapy. CADCAM can be used as the target for drug therapy, and by identifying the binding potential of CADCAM to the infected cells, treatment methods can be designed. (Kim et al. 2013).

Human-derived CRC cell lines: HCT116, LS174T and LOVO (from American Type culture collection in Rockville, MD, USA) were selected for the study. The cell lines are adenocarcinoma-derived cells under various stages of development. These cell lines resemble the environment of the human colon for Fn. Fusobacterium nucleatum nucleatum (Finn) strain ATCC25586, which can bind to CAECAM1, was used for the study.

The cell lines were grown in the T-25 flasks at 37°C in 4ml DMEM medium (Dulbecco’s Modified Eagle medium). DMEM contains 10% foetal calf serum (FCS), 100 units of Penicillin per ml of medium, 100µg/ml of Streptomycin and 2mM of glutamine.
