Isolation From The Environment. Ii First Streak

Purpose

To develop clonal purification techniques for isolating bacterial strains

To determine the CFU/ml of bacteria in a gram of soil

Procedure

In fresh plates, colonies from diverse origins and morphology were streaked from new plates. Care was taken to avoid. Ambiguous colonies, wild spreading colonies and firm colonies.

Isolation From Environment III. Second Streak

Purpose.

To develop clonal purification for isolating bacterial strains

Procedure

The method of isolating colonies from the environment was repeated in order to achieve isolate colonies. On the unchanged media, the colonies secluded were picked and restreaked.

Observation For First Streak

5.1The isolates have different morphological appearances. The isolated colonies had different shapes, color and texture. Some colonies showed a sign of mixed coloration. Some colonies, however, had similar shapes and color due to interaction with one another

5.2When multiple clonal streak isolation was done, some changes were noticed from the original colonies suggesting the presence of more than one colony. This was an indicator that the colony was not a pure colony as earlier thought. Moreover, the colonies changed their morphology due to change in constant and environmental conditions.

Isolation From Environment IV. Third Streak.

Purpose.

To develop clonal purification technique for isolating bacterial strains.

Procedure:

The same procedure as in the second streak was repeated. R2A inoculation was used to restreaked the media and then two broth tubes used to further isolate the colonies. Liquid culture from this step was kept for future rRNA sequencing

Observation For Second Streak

6.3 When the streak plate method was used to isolate the environmental colonies, the apparent purity of the isolates had different morphology in terms of size, shapes and color. There was some significant change from the previous week's result since the color of the isolations in the present week were deeper, more contrastable and the sizes had grown bigger. This means that the colonies were increasing in numbers. The general morphology was circular in shape with an entire margin. The centre of the colonies was raise to form an umbonate elevation. Moreover. The pigmentation of the colonies was yellow patches.

Storage and Preparation for Phylogenetic Characterization.

Purpose.

To determine the proper storage and preparation for phylogenetic characterization.

Procedure

A septic technique was used to maintain the purity of pure cultures.  Liquid media was the best option for culture growth. Later, the best growth from the second broth tubes was selected for future laboratory work. The centre of the stab was marked with precaution no avoid flaming sterile technique was used to keep pure cultures that were properly labelled fro future study. Finally, 0.5ml of the strains that grew best in R2Broth was provided to the TA.

Observation For Third Streak

7.2 When the isolate colonies were restreaked onto R2A, the isolates became more impure showing different colonies. The general morphology of the colonies was the large size of cells, the colonies were freely swimming on the agar plate. The colony had mas of cells organized to move it out of the main colony. This colony was different from the result of the previous week with more distinguishable colonies and massive cell outgrowth. This week’s results showed more pure colonies since the colonies used their massive cell outgrowth to move out of the mixed colonies.
